摘 要:
植物AP2/ERF是一个庞大的转录因子家族, 在植物的生长发育、器官构建及逆境胁迫和激素信号应答中发挥重要作用。本研究在茄子基因组范围内, 利用生物信息学方法对茄子AP2/ERF转录因子家族的成员、分布及结构和功能等进行分析。结果表明, 茄子AP2/ERF家族包含130个蛋白质, 其中AP2、RAV、ERF亚家族分别含有20、2和108个, ERF分为DREB/CBF (41个成员)和ERF (67个成员)两大亚族, 进化分析将二者分为11个小亚族。MEME分析表明其含有20个重要基序, 相同亚族中的ERF成员蛋白序列的氨基酸保守域构成相似, 且各亚族成员的氨基酸保守域组成类似, 表明这些基序的存在对ERF蛋白功能的执行是必需的。利用实时荧光定量PCR对茄子38个ERF基因进行组织表达分析, 结果表明其存在组织表达差异; 胁迫响应分析结果表明, 33个基因受ABA、SA、盐、重金属镉、高温和低温诱导表达, 其中部分基因显著上调或下调表达, 可能参与茄子逆境胁迫条件下的防御应答反应。本研究结果将为茄子AP2/ERF家族基因的深入研究提供依据, 为进一步解析AP2/ERF基因功能奠定基础。关键词:茄子; AP2/ERF转录因子; 生物信息学分析; 表达分析; 逆境胁迫

Abstract:
AP2/ERF transcription factors are a huge gene family in plants, and which have a variety of functions involved in developmental, organ formation, stress response, and hormonal signaling. In this study, a comprehensive computational analysis identified 130 AP2/ERF proteins in eggplant (Solanum melongena) genome, a complete overview of AP2/ERF transcription factors including chromosome locations, gene structure, phylogeny, conserved motifs and function of proteins is presented. These results showed that, AP2/ERF proteins could be classified into AP2, RAV and ERF based on phylogenetic analysis, and had 20, 2 and 108 member, respectively; 41 and 67 of them belonged to DREB/CBF and ERF gene family. The eggplant AP2/ERF proteins contained 20 important motifs based on MEME analysis, and the amino acids conservative domains of AP2/ERF members’ proteins sequence were similar in the same subtribe, and special in different subtribes members, which indicated that these motifs were necessary to the execution of AP2/ERF protein function. Real-time fluorescence quantitative PCR were used to gain insights into the function of 38 SmERF genes during various stages of development, some of the AP2/ERF genes was expressed in specific organs. 33 SmERF genes showed distinct expression patterns in response to ABA, SA, salt, cadmium chloride, high temperature and cold, the expressions of several genes were significantly up/down regulated, implying that these members might participate in regulating the defense response against abiotic stresses. These results will provide a very useful reference for cloning and functional analysis of ERF genes in eggplant.Key words: eggplant (Solanum melongena); AP2/ERF family; bioinformatics; gene expression; oxidative stress
